Why Water Content Matters in Pharmaceuticals
Moisture is one of the most critical quality attributes in pharmaceutical products. Excess water promotes microbial growth, hydrolysis of active ingredients, and physical instability. Insufficient understanding of water content leads to failed shelf-life studies, product recalls, and patient safety issues.
Karl Fischer (KF) titration, developed by Karl Fischer in 1935, remains the gold standard for pharmaceutical moisture analysis — referenced in IP, BP, and USP.
The Karl Fischer Reaction
The KF reaction involves the selective reaction of water with iodine, sulfur dioxide, a base, and an alcohol:
H₂O + I₂ + SO₂ + 3RN + ROH → 2(RNH)I + (RNH)SO₄R
The endpoint is detected electrometrically — when all water has reacted, excess iodine causes a current change that signals completion.
Volumetric KF Titration
In volumetric KF, iodine is added from a burette (either as a two-component or single-component KF reagent). The volume of reagent consumed is converted to water content.
Range: 0.01–100% water content
Best for: Liquids, solutions, high-moisture samples
Limitation: Not sensitive enough for very low moisture levels (below ~100 ppm)
Coulometric KF Titration
In coulometric KF, iodine is generated electrochemically in situ by oxidation at the anode. The amount of iodine generated (and therefore water consumed) is calculated from Faraday's law.
Range: 1 ppm – 5% water content
Best for: Low-moisture solids, APIs, excipients, oils
Advantage: Extremely sensitive; no reagent titration errors
